Human iPSC-derived Hepatic Endodermal Organoid (HEO) & Hepatic Organoid (HO)
A high-purity 3D liver organoid containing more than 98.5% ALB⁺ hepatocytes.
Comparison with Primary Human Hepatocytes (PHH)
Expression of hepatocyte markers, cytochrome P450 (key enzymes for liver metabolism and detoxification), and nuclear receptor markers was compared with PHH.
The hepatic organoid demonstrated development comparable to that of human hepatocytes.
| Cell line | IC50 (μM) | ||
|---|---|---|---|
| Benzbromarone | Diclofenac | Troglitazone | |
| HO | 163.3 | 888.2 | 122.2 |
| PHH | 111.2 | 977.0 | 171.6 |
| HepG2 | 297.6 | 1351 | 362.1 |
Cytotoxicity assays were conducted on hepatic organoids (HO), primary human hepatocytes (PHH), and HepG2 (liver cancer cell line).
Results showed that HO exhibits drug toxicity responses more similar to PHH, with a clear distinction from HepG2.
CYP450 Enzyme Activity in Hepatic Organoids
CYP450 enzyme activity was evaluated directly in hepatic organoids.
The activity of several key CYP450 enzymes was confirmed through their specific metabolites.
